玩家在選擇角色後再挑賽道。可以使用道具攻擊其他玩家，目的在比賽中獲得最佳名次。支持單人模式和多人模式，多人模式中最多有兩位玩家同時進行比賽。

操作相對簡單，使用超任的控制器來操縱賽車跟轉向。B:加速、Y:剎車、A:使用特殊道具、Start:暫停或選擇、Select:切換視圖、L:漂浮、R:跳躍。

遊戲共有20個不同的賽車跑道，又分為四個不同的盃賽（Cups），有蘑菇盃（Mushroom Cup）、花盃（Flower Cup）、星盃（Star Cup）、特別盃（Special Cup）。

每個盃賽有五個不同的賽道。各個賽道都有其獨特的設計和挑戰，包括彎道、障礙物和特殊地形。可供單人或多人遊玩。沒有傳統的頭目出現，遊戲的重點是在單人或多人模式中與其他車手競爭。畫面使用強制分屏視圖，提供精彩的賽車體驗。

主要密技：

解鎖:在日版的計時賽(Time Trial)或一對一的比賽模式(Match Race)，看到盃賽Menu畫面時，輸入L、R、L、R、L、L、R、R、A，如果操作正確就會聽到Boo的聲音，這時候特殊盃賽就會解鎖。

縮小:來到角色挑選單，按住Y再按A，角色就會在比賽時縮小，對車速沒有任何影響，再同樣操作一次就能變回正常。

互換畫面:用2P的手把，按住L跟R直到選角畫面出現。開始比賽的時候，發現螢幕已經互換。

挑戰失敗後繼續:在對戰模式(Battle Mode)中，只剩下一個氣球的時候，跳過一個香蕉皮，音樂會改變並且螢幕變暗，這時候仍可以持續開卡丁車。當拿到物品的時候，無敵星星的音樂就會響起。

刪除存檔:如果想要刪除存檔，可以在標題畫面暗L、R、Y、A即可。如果是計時賽，在賽跑紀錄畫面進行一樣的操作即可。

跳過車道:在一些關卡只要透過正確的使用羽毛或密技，可以略過部分跑道。關卡有庫巴城堡(Bower's Castle Courses)、幽靈谷(Ghost Vallley Courses)、多軌道(Multiple Tracks)、彩虹路(Rainbow Road)、香草島(Vanilla Island Courses)。

增加台數:若三場完成的賽事排名都一樣，就能增加一台數。

加速:當比賽準備正式開始，在第二個燈亮起來前，按B(加速紐)直到開賽。如果控制得宜，很容易就得到第一名。

選擇車手:進行單人的計時賽，在選擇角色的畫面中用2P的手把按開始鍵，這時候會出現電腦的對手選項，可選隱藏的幽靈角色。

攝影機搖晃:在計時賽進行時都沒有失誤(沒有掉落或是離開跑道)，當看到重播的比賽影片時候，按L或R就能讓攝影機搖晃。

1.單人模式：

設法在有限的時間內狠狠甩開對手，克服所有道路上的障礙後迎向最後勝利。螢幕的上半部分顯示卡丁車後視圖，下半部分則可以切換為後視鏡視角或全局俯視圖，顯示所有車手的位置。這使玩家能夠掌握整個賽道的局勢。

瑪莉歐GP（大獎賽）：

這是主要單人遊戲模式，玩家將與七名電腦控制的車手競爭。遊戲提供不同的動力等級，包括50cc、100cc和150cc，但要解鎖150cc，必須在100cc的難度下完成遊戲。

越高等級會面臨更困難的車速競賽。特別盃需要在100cc模式下贏得其他三座盃賽的金牌才能解鎖。必須以第四名或更好的成績完成比賽才能晉級下一個賽道。最後一名的車手被淘汰，在比賽過程中可以選擇繼續嘗試或退出。

計時賽:在100動力級進行，沒有任何道具的狀況下，設法用最快的速度完成五圈，最終會結算出每個賽道使用的累積時間。因為超任主機的限制，所有時間紀錄在關機後就會自動被刪除。如果整場賽事都沒有發生掉落或是離開跑道的情事，下次的賽事就會出現幽靈，Switch的線上移植版也有同樣的設定。

2.多人遊戲模式：

瑪莉歐GP（大獎賽）：類似單人遊戲模式，但少一名電腦車手。想要挑戰下個賽道，至少一位玩家的排名要在前四位。如果選手連續三次都沒有排名，則遊戲結束。

比賽模式(Match Race/VS. Mode)：兩名玩家進行一對一的比賽。

對戰模式(Battle Mode)：在戰鬥模式中，兩名玩家可以挑選四個特別設計的場地進行，用香蕉皮或庫巴殼來擊破其他車手的三個氣球，擁有最多氣球的人獲勝。
